Trace Fluorescence Imaging
Trace Fluorescence is mainly used when the intrinsic protein fluorescence is too weak to be seen and becomes noisy or the camera shutter time exceeds an acceptable interval. In this case, the used fluorophore was carboxyrhodamine. An advantage of carboxyrhodamine is that its fluorescence can be excited at a maximum of 525 nm (green light), which reduces the radiation dose to the crystal significantly. Moreover, the background fluorescence is often reduced since most plate materials do not show any fluorescence at this wavelength. Once the covalent staining protocol is established, the procedure is simple and can be applied routinely.
